银杏雌雄株在干旱胁迫下转录组的比较分析毕业论文

摘 要

雌雄异株植物占被子植物总种数的4%~10%。在全球变暖局势下,干旱对植物生长的影响愈发明显,而雌雄异株植物的雌株和雄株的生理生化变化在干旱环境中,因为结构、职能等的不同而有所不同,因此对干旱条件下雌雄异株植物的研究就显得格外重要。

目前关于抗旱基因的研究大多是从模式植物如拟南芥中进行,而木本植物的抗旱性研究大多停留在生理指标层面,对于基因方面的研究比较单一且比较缺乏。银杏作为比较古老的雌雄异株植物,有着基因组大、对非生物和生物胁迫抗性强、雌雄异株繁殖等一系列吸引人的特点,是生物学研究的理想模式种,然而,由于缺乏高质量的基因组序列,理解它的生物学和进化存在着不小的障碍。

本研究共完成了银杏24个样品的转录组分析,共获得171.65GB有效数据,各样品有效数据均达到 6.01GB,Q30碱基百分比在88.70%及以上。分别将各样品的有效数据与指定的参考基因组进行序列进行了比对,比对效率从73.27%到82.25%不等。本论文选取了7月24日和同年9月7日时期的银杏植株样本的数据进行撰写,得知了干旱胁迫下基因表达差异最大的五个通路(碳代谢、氨基酸生物合成、内质网蛋白质加工、植物激素信号转导、淀粉蔗糖代谢),雄性银杏植株在干旱胁迫下比雌性植株更敏感,雌性植株对干旱的适应性要大于雄性。

Comparative analysis of transcripts of female and male Ginkgo biloba plants under drought stress

ABSTRACT

Androgynous plants account for 4% ≤ 10% of the total number of angiosperms. Under the situation of global warming, the effect of drought on plant growth is becoming more and more obvious, while the physiological and biochemical changes of female and male plants in arid environment are different because of their structure and functions. Therefore, the study of androgynous plants under drought conditions is particularly important.

At present, most of the studies on drought resistance genes are carried out from model plants such as Arabidopsis thaliana, while most of the studies on drought resistance of woody plants stay at the level of physiological indexes, and the research on genes is relatively single and scarce. Ginkgo biloba, as an ancient androgynous plant, has a series of attractive characteristics, such as large genome, strong resistance to abiotic and biological stress, androgynous reproduction and so on. It is an ideal model species for biological research. However, Due to the lack of high quality genome sequences, there are many obstacles to understanding its biology and evolution.

In this study, the transcriptional group analysis of 24 samples of Ginkgo biloba was completed, and the effective data of 171.65GB were obtained. The effective data of each sample reached 6.01GB, and the percentage of Q30 base was 88.70% and above. The effective data of each sample were compared with the specified reference genome sequences, and the comparison efficiency varied from 73.27% to 82.25%. In this paper, the data of Ginkgo biloba plant samples from July 24 and September 7 of the same year were selected to write. It was found that the five pathways with the greatest difference in gene expression under drought stress (carbon metabolism, amino acid biosynthesis, endoplasmic reticulum protein processing, plant hormone signal transduction, starch sucrose metabolism). Male ginkgo biloba plants are more sensitive than female plants under drought stress, and female plants are more adaptable to drought than males.

1 前言

1.1 研究背景

地球上有52.2%大陆属于干旱与半干旱地区,而湿润与半湿润的沿海季风区有时也会经历阶段性的干旱胁迫。水是生命之源,对植物而言,水有着重要意义。

植物的一切生命活动只有在一定的水分条件下才能进行。主要表现有:(1)水分是细胞质的主要成分。细胞质含水量在70%~90%,使得细胞质呈溶胶状态,从而保证了代谢活动的正常进行,如果含水量减少,则细胞质会变成凝胶状态,从而大大减弱了生命活动。(2)水是代谢作用过程中的反应物质。在光合作用、呼吸作用、有机物质合成和分解过程中,都有水分子的参与,如果受到干旱胁迫,这些代谢作用将会收到影响,从而影响植物正常的生长。(3)水分是植物对物质吸收和运输的介质。一般情况下,固态的无机物质和有机物质不能被植物直接吸收,只有溶解在水中才能被吸收。而各种物质的运输也需要溶解在水中才能进行。(4)水分能使植物保持姿态。由于细胞含有大量水分,使细胞的紧张度得以维持,从而使得植物枝叶挺立,便于充分接受光照和进行气体交换,也使花瓣张开,有利于传粉。(5)水分在植物的生态环境中也有重要作用。水可以调节植物体温,保证温度相对稳定;能调节植物周围的环境,增加湿度,改善土壤及其表面温度等。

因此,干旱对于植物生长和作物的产量都会产生不利的影响。缺水会阻碍植物的营养生长,限制植株的伸长生长,使植株总叶面积减小,会使呼吸作用、光合作用下降,影响植株体内的各种生理生化进程,影响植物生殖器官分化及发育,进而影响植株的生殖生长,加速植物衰老[23]
