黄颊长臂猿微卫星DNA遗传标记的筛选及在亲子鉴定中的应用毕业论文

摘 要

黄颊长臂猿属于冠长臂猿属(Nomascus)的黄颊长臂猿种(Nomascus gabriellae),属于国际自然保护联盟(IUCN)红色名录中濒危(EN)物种。本研究采取微卫星技术对红山动物园内的14只黄颊长臂猿进行基因分析,同时基于本研究扩增测序结果分析显示,本研究共获得11个多态微卫星位点,等位基因为2~5个。多态性信息含量范围PIC为0.175~0.701,观察杂合度(Ho)为0.067~0.8,期望杂合度(He)为0.191~0.770。两个位点与Hardy-Weinberg平衡有很大的偏差(平衡(Plt;0.01)。这些微卫星位点的研究结果阐述了近缘物种SSR在黄颊长臂猿中的通用性,揭示南京红山动物园内14只黄颊长臂猿的遗传变异及亲缘关系。同时对比对应园内已知的亲缘图谱和亲缘系数,显示得到的微卫星位点符合科学的实验结果,可应用于黄颊长臂猿种群的管理和保护,为保护黄颊长臂猿提供理论基础。推动圈养长臂猿种群从注重数量增长步入种群数量及质量同步发展的良性循环之中,对挽救黄颊长臂猿种群有一定意义。

Screening of microsatellite DNA genetic markers of red-cheeked gibbon and their application in paternity identification

ABSTRACT

The red-cheeked gibbon belongs to Nomascus gabriellae, an endangered species in the Red List of the International Union for Conservation of Nature (IUCN). In this study, 14 red-cheeked Gibbons in Hongshan Zoo were analyzed by microsatellite technology. Based on the results of amplification and sequencing, 11 polymorphic microsatellite loci with 2-5 alleles were obtained. The polymorphic information content ranged from 0.175 to 0.701, observed heterozygosity (HO) from 0.067 to 0.8, and expected heterozygosity (He) from 0.191 to 0.770. There was a large deviation between the two loci and Hardy-Weinberg equilibrium (P lt; 0.01). The research results of these microsatellite loci illustrate the universality of SSR in red-cheeked gibbons, reveal the genetic variation and relationship of 14 red-cheeked Gibbons in Hongshan Zoo, Nanjing, and the correlation coefficients with the known relative maps in the zoo. It is proved that the obtained microsatellite loci conform to the scientific experimental results and can be applied to the management and protection of red-cheeked gibbon population. To provide a theoretical basis for the protection of the red-cheeked gibbon. Promoting captive gibbon population from focusing on quantity growth into a virtuous circle of simultaneous development of population quantity and quality is of certain significance for saving the population of red-cheeked gibbon.
