摘 要

繁殖模式由卵生向卵胎生转变是重要的进化事件。卵胎生进化过程中逐渐延长卵滞留的时间使卵获取在母体内更长的发育时间,其选择利益和面临的主要限制因素是学术界普遍关注且尚待解答的重要科学问题,但相关研究却十分匮乏,迫切需要选取合适的动物模型开展深入研究。一般认为,生理制约是影响物种从卵生向卵胎生进化过程中的主要限制因素,而母体内的低氧环境是其中重要的生理制约因素,是卵胎生进化过程中需要克服的难题之一。本实验以荒漠沙蜥为材料,通过控制卵在不同的氧浓度环境下孵化,比较不同氧浓度下各胚胎时期发育速率、新生卵的孵化成功率和孵出幼体的表型及适合度。检测氧气可得性对卵滞留的制约作用。研究结果旨在探讨爬行动物卵滞留时间、生理制约机制和进化选择利益,为阐述卵胎生的渐进式进化提供实验证据。

Influence of oxygen availability on egg incubation and hatching phenotypes of Phrynocephalus przewalskii

ABSTRACT

The evolution of reptilian viviparity (live bearing) from oviparity (egg laying) is a nature’s great event. Longer periods of egg retention by the gravid female is thought to require transitional stages of increasingly longer periods of embryonic development in utero. The selective advantages and constraints associated with egg retention arouses pervasive focuses in academic circle, however, correlational research is still deficiency,and the urgent need is to carry out in-depth study with proper animal mode. It is generally acknowledged that physiological constrain is the main limiting factor, while hypoxia (oxygen deprivation) is the primary physiological factor that limits development of embryos in the oviducts. The objective of this research was to test the hypothesis that the rate and degree of development that reptilian embryos undergo in utero and after oviposition is related to the amount of O2 available to embryos, by incubating eggs of Phrynocephalus przewalskii under conditions of low, normal and high oxygen. Moreover, Squamate reptiles provide a very useful model system for studying the evolutionary transition from oviparity to viviparity because they exhibit a wide reproductive diversity well beyond a simple oviparity/viviparity dichotomy and because viviparity has evolved far more often in this group of animals than in all other non-mammalian vertebrates combined.

1 引言

繁殖是一个生命个体产生另一个新的生命个体的生理过程,是基本的生命特征之一。根据达尔文进化论,物种按由低级到高级,由简单到复杂的进化趋势进化;同样地,动物的繁殖模式也存在着同样地规律。由简单、低级的卵生繁殖模式进化成为复杂、高级的卵胎生繁殖模式,并进一步进化成更复杂、更高级的胎生繁殖模式。运用系统发生分析显示在脊椎动物进化过程中至少有141次独立的卵胎生进化起源[1],其中爬行类占到~115次[2]。因此,爬行类在卵胎生进化起源方面占绝对优势。同时,爬行动物(蜥蜴、蛇和蚓蜥)有卵生、卵胎生和卵生-卵胎生三种繁殖模式,是研究卵胎生进化理想的实验动物。

研究者们认为由卵生进化到卵胎生是渐进式的,即:卵生种通过逐渐延长卵在母体内的滞留时间,而逐渐进化成卵胎生的繁殖模式[3]。而卵滞留的选择利益和面临的主要限制因素是学术界普遍关注且尚待解决的重要科学问题。其中,制约因素主要有系统进化方面、生理方面和生态方面上的制约,如气体交换、温度、湿度等。而一般认为,生理制约在影响物种从卵生向卵胎生进化的过程中起到的制约作用是最重要的,即使生理制约并不是卵胎生进化的唯一制约条件。其中,母体子宫中的氧气不再能够满足胚胎对氧气日益增长的需求是使高度滞留卵生长减慢或停滞的主要原因[4]

本实验以荒漠沙蜥为材料,通过控制卵在不同的氧浓度环境下孵化,比较不同氧可得性下各胚胎时期发育速率、新生卵的孵化成功率和孵出幼体的表型及适合度。检测氧气可得性对卵滞留的制约作用。研究结果旨在探讨爬行动物卵滞留时间的表型可塑性、生理制约机制和进化选择利益,为阐述卵胎生的渐进式进化提供实验证据。值得注意的是,在现有的相关研究中,几乎都是以卵胎生种类作为研究材料,但卵胎生种类正是处于卵生卵胎生连续谱的卵胎生终端,因此无法为该繁殖模式渐进进化的过程提供有力的线索,所以需要从现生卵生种类中寻找有关证据,而相关研究仍十分匮乏。从现生卵生物种寻找卵胎生渐进进化的证据是本项研究选材的特色。同时,在不同氧气浓度下孵化新生卵,检测胚胎发育速率、孵化成功率和后代适合度的组间差异,能直接检验氧浓度对卵滞留的生理制约作用。
